A number of factors have been proposed to explain beta-cell failure. These include individual differences in beta-cell mass, aging and primary beta-cell defects. The process of aging is associated with a decrease in beta-cell number. It is also likely that in some people there are genetic determinants of beta-cell abnormality affecting either insulin processing or the signalling pathways that lead to impaired insulin secretion.
