Activation of D2 receptor inhibits adenylate cyclase and, decreases of cAMP, also – activates K+ channels and inhibit polyphosphoinositide turnover.

D1 are most abundant in striatum, limbic system, thalamus and hypothalamus,

D2 - in striatum, limbic system, thalamus and hypothalamus and pituitary gland,

D3 – in limbic system, but not in the striatum,

D4- is much more weakly expressed, mainly in the cortex and limbic system.

The functions of DA-ergic pathways divide into:

- Motor control ( nigrostriatal system)

- Behavioral effects (mesolimbic and mesocortical system)

- Endocrine control (tuberohypophyseal system).
