The next step in our experiments is to conduct the TLI/ATG conditioning regimen on mice affected with EAE. To induce disease, we inject the mice with an emulsification of myelin oligodendrocyte protein (MOG) in Complete Freund’s adjuvant, along with a dose of pertussis toxin. 2 days later, we give a boost of pertussis toxin, and 12-14 days later, EAE develops in approximately 80-90% of susceptible animals. At the bottom of the slide are spinal cord sections. On the left is a normal spinal cord from an unimmunized animal, and on the right is the inflammation and destruction of myelin by activated T cells evidenced in a MOG-immunized animal.
