MOVIE DESCRIPTIONS for QuickTime movies of HK97 Capsid Expansion
OUTSIDE.MPG and INSIDE.MPG were the first movies of viral capsid maturation. They were included as Supplemental Data to http://www.cell.com/cgi/content/full/100/2/253/DC1.
OUTSIDE.MPG Mpeg Video (4.4 MB) Dynamic visualization of maturation ("expansion") of bacteriophage HK97 capsid, as induced in vitro by acidification. The icosahedral capsid is viewed from the outside along a 2-fold symmetry axis. The video is based on cryo-EM 3D reconstructions at ~25 Angstrom resolution of the initial state (Prohead II), final state (Head), and three transitional states--Expansion Intermediates I, II, and III. When expansion is induced in vitro by switching to pH 4.18, the three intermediates are abundant at 5, 37, and 130 min, respectively (labels, upper right). The experimental images of these conformational states are connected by morphing. Thus, the movie does not represent the acid-induced expansion event in real time, but may more closely emulate the time course of capsid expansion in vivo.
